How soon after it comes out should I be able to get it somewhere? I haven't pre-ordered it, and am not in a huge rush to have it, but I'm sure once I hear about it day in and day out I'll want to go pick it up and dive in.

I would assume you could get it at best buy on release day....
From my experience and from other friends experience, they rarely sell out of games, even for huge releases.
There were plenty of Halo: Reach copies at BB on release day. FWIW
