www.childrensmuseums.org will have a list. You can check any cities you know you will or would like to hit. Also check out the Reciprocal Network.
www.astc.org They have a list of their membership and details of the reciprocal benefits each offers.
Some places refer to themselves as children's museums but are affiliated with ASTC. Some science museums accept children's museums memberships. Sometimes a facility with accept either. We live in Mississippi and belong to the natural science museum in Jackson, which last year was $65 for our family of 7. We went to McWane Science Center in Birmingham, a phenomenal place fun for kids and adults alike, at no charge.
On occasion we haven't planned ahead and just used our GPS to tell us what was coming up along the way. We stopped in Shreveport on our way back from riding the Polar Express in Texas about six years ago and enjoyed an impromptu stop at that science center.

Nashville and Louisville are the best and most interesting cities on your route. Cincinnati isn't bad in parts, and I like Memphis for what it is, but I probably wouldn't want to be taking a family through an extended trip there. Columbus is fine and family-friendly, but it's as suburban and stale as anything I've seen. Cleveland is really cool in parts, but it's probably not worth the headache from Zanesville.
It really depends on what you want to do. Stuff for kids? Nice restaurants? Shows? More natural stuff?

Mammoth Cave for sure then. I'd almost say that's a must with the route you're probably taking.
Lots of stuff to do in Nashville...others would probably be better qualified to comment on that.
Louisville Slugger and Muhammad Ali museums in Louisville. Churchill Downs and the Kentucky Derby Museum. If you're a bourbon guy, Makers Mark, Heaven Hill, Four Roses, and Jim Beam are near I-65 just south of Louisville.
Red River Gorge is great hiking outside of Lexington (would be a bit of a detour).
King's Island outside of Cincinnati is great for kids. The Newport Aquarium (the Cincy suburbs on the KY side) is very cool for kids. You could catch a ballgame at Great America - it's a pretty damn cool place to catch a game, and the Reds have been good for the last three or four years, and it's not too expensive.
Cedar Point in Sandusky, Ohio is one of the best amusement parks in America, but it's also a bit of a detour (couple hours) from your route and final destination. That might be worth checking out.

Thanks for everyone for telling me how terrible my state is, I finally see the light. OP, if you come through Columbus you should check out the Columbus Zoo, you could also check out a Columbus Clippers' game. Those are two things your kids would enjoy. Also Ohio Caverns is a very unique spot northwest of Columbus.
