I think a reboot would have to largely involve a new crew. The people pictured with Fillion would be the most likely to not return. I think Jayne and Inara would have moved on, and while it might make sense for River and her brother to be with them still, having a weapon as powerful as River could ruin many story lines. There's also the Summer Glau Curse saying don't bring her back. To me it'd be Mal, Zoe, maybe Kaylee and everyone else would have to be new.
