Forget a hotel, rent a house. If your group is not too big, hire a captain to take you out on a booze/fishing cruise to the sand bars around the island. Much better and more personal than one of those big tourist boats. Also check out Dante's for the pool party, and Bare Assets for strippers.

Key West (the town) during the day is pretty boring. You want to be on the water.
