Here's a good opener I stole from the interwebs:
"Joe had a hard time choosing his best man for this day. First he called up his most charming friend, and he said, "Sorry, Joe, I can't." Next he called up his funniest friend and he said, "Sorry, Joe, I can't." Then he called up his most handsome friend and he said, "Sorry, Joe, I can't."
Finally, he called me. And I said, "Joe... I can't say no to you four times."

The trick is not to make it about you, but about them. That is somewhat clever, but you can come up with something clever that involves the couple. IT's not supposed to be a standup routine by the best man, tho of course a laugh is a good thing.
