Mammoth Cave for sure then. I'd almost say that's a must with the route you're probably taking.
Lots of stuff to do in Nashville...others would probably be better qualified to comment on that.
Louisville Slugger and Muhammad Ali museums in Louisville. Churchill Downs and the Kentucky Derby Museum. If you're a bourbon guy, Makers Mark, Heaven Hill, Four Roses, and Jim Beam are near I-65 just south of Louisville.
Red River Gorge is great hiking outside of Lexington (would be a bit of a detour).
King's Island outside of Cincinnati is great for kids. The Newport Aquarium (the Cincy suburbs on the KY side) is very cool for kids. You could catch a ballgame at Great America - it's a pretty damn cool place to catch a game, and the Reds have been good for the last three or four years, and it's not too expensive.
Cedar Point in Sandusky, Ohio is one of the best amusement parks in America, but it's also a bit of a detour (couple hours) from your route and final destination. That might be worth checking out.
