It seems like the most comfortable controller would be the 360 but put the right thumbstick where the A,B,X,Y buttons are and bring those buttons down to where that thumbstick is now.
Would have both thumbs in same position and relaxed. Wonder what that would be like.

The reason the joysticks on the 360 controller are in the optimal position is because the left joystick, being for character movement, is positioned perfectly for vertical, forward and backward, motion. Your left thumb only has to flex or extend to move forward or backward.
The right joystick is placed for horizontal rotation of your vision and it's placement allows for left or right movement through only flexion or extension of your right thumb.
It's designed to allow for the widest range of motion with the lowest energy exertion. It's anatomically correct.
The PS controller is not. The right joystick is in the correct position, but the left is not.
