I don't think he knows. Vargo doesn't know in the books, but cuts Jamie's hand anyway. When he finds out that he is going back to Kings Landing, he gets worried that Tywin will be mad.

I think, just speculation on my part, that Locke thinks they will be sending an alive Jamie back to Riverrun (sans hand).

I think in the books Vergo's chopping of Jaime's hand made perfect strategic sense - don't recall the details - but I recall Roose showing the logic how it wasn't just a random/crazy thing, but a well thought out strategic move.
