Perhaps he wasn’t the main villain of Ferris Bueller’s Day Off, however, the maître D at Chez Quis should get some consideration for Supporting Villain in a Comedy...

Most of the classics have been mentioned but here are a few that haven't. Carrie Fisher in The Blues Brothers, Chloris Leachman in Mel Brooks High Anxiety, and Spencer Tracy as Culpepper in It's a Mad, Mad, Mad, Mad World.
