I'm busy being unproductive at work while browsing Amazon when I realized how many great games I haven't played. I missed a lot of shite during the year or so I took off from gaming. So help me choose! Below are the games I've had some interest in playing at some point but never got around to them.
Uncharted 1-3
Mass Effect 1-3
BioShock Infinite (loved the first one)
Dead Space 1-3
Skyrim (played the shite out of Obilivion)
Borderlands 1 & 2
Infamous 1 & 2
Fallout: New Vegas
Dishonored
Sleeping Dogs
Uncharted 1-3
Mass Effect 1-3
BioShock Infinite (loved the first one)

Out of that list, my top three would be:
Bioshock Infinite
Uncharted 1-3
Dead Space 1-3
I threw Dead Space in as it and Sleeping Dogs branch out of a bit from just shooting.

I haven't played all of these, but they are all good games with loads of endorsements from various game review sites, so I don't think you'd go wrong picking any of them.
What sort of games are you in the mood for?
Without knowing that, if I had to pick just one, I'd probably say Uncharted 2. As I've said in other posts, I think it's one of the best games this gen.

I really enjoyed the dead space series. Im probably alone when I say that. It has a really good story and a bit of horror. There were some moments in the first 2 where I was on the edge of my seat waiting for something to scare the ahit out of me.

I think eventually I'm going to give Dead Space a shot. I played the demo. Seemed pretty difficult, but that was 5 minutes of gameplay

I couldnt put it down when I started. The first wws the best. The second was on par with the first and 3 was a but too hollywood but had some scarier foes. If you enjoyed resident evil type horror give it a shot

Uncharted 2 and Mass Effect 2 are absolutely unmissable.
You don't really need to play the first games in the respective series, either.

Are you on PC?
RPGs
New Vegas and Skyrim fully modded are amazing and will provide as many hours of play as you want. Side note, if you haven't played FO3, it's a better game than New Vegas IMO.
The Mass Effect series, despite the conclusion, is one of the best gaming trilogies ever. So that definitely deserves a look.
Aside from those three games I would only recommend Dishonored if you want a great stealth-based action adventure game and Bioshock Infinite if you want a great shooter with a great story and narrative.
So, it comes down to your preferences.

Overall rating, IMO:

My little brother used to play the shite out of Dynasty Warriors when he was a kid. He has probably orphaned hundreds of thousands Chinese children.
