I only heard of this the other day.
Forewarned... it was in the context of "The bourbon trail, for some unknown reason, is extremely popular with gay men."
And no. I am not joking.

Only have done makers mark so no help on others. But was a good time.

We did this 2 years ago and had a great time. We went to Makers, Wild Turkey and Woodford Reserve. As far as the distilleries go, Makers probably has the best tour/experience but Woodford is on an old horse farm and probably has the best scenery (if you're into that sort of thing).

A friend of mine did the trail last year. He visited Beam, Heaven Hill, Makers, Four Roses, Turkey and Woodford in two days. He said it was fun and the Maker's was the best hands down...

I have visited Jack Daniels. Very nice place, but no tasting because it is dry and there are no other distilleries around. A long weekend in Louisville with lots of tours and a place to party at night would make for a good time…

I visited the Jack Daniels Distillery last year while turkey hunting in TN. We actually hunted in the shadows of one of the many barrel houses in the area. Its a good place to tour and a neat operation. No tasting other than a complementary lemonade after the tour...

I have been to Buffalo Trace, Woodford, and Makers
I liked Buffalo Trace as the best all around tour/facility/info. Woodford was the nicest. Makers probably had the best tour

Makers was hands down the best in terms of the attention to detail in their tour. You can tell that invest a lot of time and effort into trying to draw the consumer in, and it was largely successful. Buffalo Trace was extremely disappointing for me. Woodford was a solid second.
We stayed in Bardstown, which wasn't bad. Kinda central. Makers is way off the beaten path, but the hotel was decent, super cheap, and we were able to get practically anywhere we wanted to with few difficulties. Keep in mind that outside of the distilleries the place is pretty slow, so you should plan ahead to make sure you don't have too much idle time or else you'll be bored as all get out. Enjoy!

I did it almost two years ago, though my experience wasn't the same as you will have. We did a 200 mile relay run called the Bourbon Chase where we ran from distillery to distillery over the course of two days. Didn't have time to take tours, but we had time to take in the scenery and a quite a few sips of whiskey.
Makers and Woodford were the best, imo. Makers is out in BFE, but worth the hike. Reminded me of the JD distillery as it it's up in a holler and has a creek running through it. Woodford was cool because of the ol' time feel with the barrel tracks and whatnot, plus it has some amazing thoroughbred farms around it. Heaven Hill has a nice museum, but not very much else. Jim Beam, Four Roses, and Wild Turkey were meh.

Makers for the sake of notoriety and a nice setup. Woodford is just a nice tour. You really can't go wrong with the major distilleries IMO.
While you're around Lexington or Louisville you should check Keeneland and Churchill Downs for racing. Always a good experience.
If you don't mind spending the cash and it's not too terribly close to Derby, stay in Louisville at 21c Museum Hotel and drop in Proof on Main for some fantastic area dining. If it is, you may want to check out some Bed and Breakfast setups in Bardstown, KY.
