What book would it be?
Mine would be lord of the flies. If I had enough talent and money this would definitely be my passion project.

The Lock Artist by Steve Hamilton
It actually has a lot in common with Drive. The big difference is that the lead character is mute in The Lock Artist but you can certainly work your way around that problem with voice-over narration and other creative techniques.

Juliet, Naked - Nick Hornby
High Fidelity is my favorite novel/film combo, and I think that Juliet is on the same quality level as Fidelity. Could be a great film if Hornby gets involved with the script and production.

For novels that haven't been made or are about to be made into film I'd say Altered Carbon by Richard Morgan. Zelazny's Amber books as mentioned would be pretty awesome as well. The Lies of Locke Lamora would make an excellent film as well.
For TV I'd really like to see the Dresden Files done well on HBO.
