I worked at Disney World for a small amount of time.
-Navigate the parks in the opposite direction as most people. (Ie: in Magic Kingdom, most people start out by going to Main Street, then Tomorrowland, then around counter-clockwise the whole time).
-Don't forget that FastPasses now expire if you don't use them within the allotted time frame. This is a brand new policy, and many are not aware of it.
-If you have not already, download the DisneyMagic app for your smartphone. It will tell you wait times for all of the rides, where various characters are located and what times they will be there. It is a HUGE time saver.
-If you plan on riding any big ticket rides (Rockn' Rollercoaster, Space Mountain, Splash Mountain, Everest, etc. Send either you or your wife to go grab a fast pass for that ride immediately as soon as you enter the park. The fast passes for those rides are typically all gone within a few hours and you will then be stuck waiting in a 2 hour line.
-Don't forget about rider switch passes! Seeing as how you have small children. If you and your wife still want to ride a big ride that might be too much for them, go to the front of the line and ask for a rider switch pass. You will stand in line and ride the ride while your wife waits with the kiddos, then when you are done, give the pass to your wife and she will get to cut to the front of the line (or at least go through the fast pass line) and ride while you wait with the kids.
