I understand Land Rover stopped selling the Defenders in the US about two decades ago. But is there a way to still get one?

It is not legal due to emissions standards if I had to guess
Therefore you probably can't get it registered

You CAN buy any vehicle you can afford to ship over and pay import duties on. Unfortunately the Defender is not DOT certified. You cannot drive it on US streets. Now there are ways around this if you can afford it. You have to get the vehicle shipped over in parts with no engine or drivetrain. You then have to buy an engine and drivetrain from a US land Rover parts supplier. You then have it built together and you certify it as a kit car. It will be licensed and insured as such. This is the only real way around it.
a lot of people were bringing in Japanese spec cars and defenders and minis, and the Government has really been cracking down on this and destroying them.

Go to a junkyard. Plenty to choose from.

What is preventing them from being DOT certified when they were at one point?

They don't meet new requirements.
Safety, emissions and such. Plus the ban on importing.

As it currently stands in the U.S., it is generally not legal to import a car from another country unless that car is more than 25 years old. There are a few exceptions to this, like the show and display rule, but generally this law has kept a lot of great machines out of the hands of American enthusiasts.
The stated reasons for this are to keep cars not certified by U.S. safety standards and emissions laws off our roads, but it's really to protect the interests of automakers who don't want you buying the cars they sell abroad when you can buy the cars they have in U.S. showrooms instead.
