I could listen to In Memory of Elizabeth Reed from Live at Filmore East a repeat forever and likely always find something new in the song I had never noticed before. Love it.

The greatest song ever is, in my opinion, either "A Day in the Life" or "Us and Them." "Comfortably Numb" enters into the discussion as well.
The greatest piece of music ever is a separate and much more inclusive question. There are two pieces of music that give me chills every time I hear them, no matter how often I hear them. One is the Tannhauser Overture by Wagner. The other... there is a legend that, one day, one of Handel's servants entered the study to find the composer crying at his desk. He rushed to his master's side and asked him what had caused him to weep, and Handel told him, "I had a vision in which the heavens were opened to me, and I saw God seated on His throne." He had just finished composing the Hallelujah Chorus.

Whole Lotta Love--Led Zeppelin
I love the entire song. It is a never skip, even with the radio play. +1 for Achilles Last Stand
Cruisin Together -- Smokey Robinson
I don't know if there is a smoother song. It's just excellent
Smells Like Teen Spirit -- Nirvana
Kind of a cliche'd pick, but it's impact certainly changed the landscape. It isn't close to my favorite Nirvana song, but it certainly fits the mold.
Suspicious Minds -- Elvis
There isn't much that needs to be said. Dude could just perform and the song is fantastic.
