While everyone has suggested wonderful sandwiches, I'm going to offer one I had last week that was on the lighter, "girlier" side.
We stopped on Magazine to get a snack from La Boulangerie. My wife grabbed a La Fromager, and it was absolutely incredible. Triple creme brie, tomatoes, roasted red peppers, lettuce, and ciabatta (I think).
I wouldn't choose it over a shrimp, roast beef, or oyster po boy nor would it come close to a muffalatta, but in the realm of cafe-style sandwiches, it was one of the best I've had.

BBQ shrimp po-boy - Liuzza's by the Track (New Orleans)
Le Pig Mac - Cochon Butcher (New Orleans)
hot sausage po-boy - Sammy's (New Orleans)
hot sausage po-boy - Gene's Po-Boy's (New Orleans)
fried shrimp Po-Boy - Olde Time Grocery (Lafayette)
Parrain's Special (grilled cheese with boudin) - Johnson's Boucanière (Lafayette)
The Single - The Company Burger (New Orleans)
double cheeseburger - Judice Inn (Lafayette)
roast beef po-boy - R&O's (Metairie)
roast beef po-boy - Sammy's (New Orleans)
Ray Ray (fried chicken breast with ham and Swiss) - Sammy's (New Orleans)
bahn mi - Tan Dinh (Gretna)
Really, just about anything from Sammy's and Cochon Butcher could make the list.
The fried shrimp po-boy from Parkway would make the list, except that none of the five times I've had it has the shrimp been truly hot. The quality of the shrimp is excellent and it's always cooked just right. But after so many times I have to acknowledge that it's never hot, never cooked (or at least served) to order. Too bad.
